Disabling an pen drive is simply opposite of enabling your pen drive. For disabling your pen drive you have two option one is to first go to Device manager and click on USB controller and then right click on USB mass storage and click on disable to perform this task. And the second way is, Goto registry editor by typing regedit in RUN and in registry editor find this addess HKEY_LOCAL_MACHINE\SYSTEM\CurrentControlSet\Services\UsbStor
now at this address you will find an option of start, right click on start and click on modify and now change the value by 4 to disable you pen drive.
